Thinking of visiting WDW for one day in early June. It will be the first time for my kids (and me), a 6 yr old boy and a 2 yr old girl. What would be the best things to do/see to make the most of just one day but not overdo it!

One day!? Not a problem. I would go to Magic Kingdom as this is truly what guests think of first when you say Walt Disney World.
I would ask my pirate what three things he wants to do. This way he has a say and since you will not see everything, he gets his check list done. You may need to eliminate some of the bigger rides (Space Mountain, Splash Mountain, Big Thunder Mountain Railroad and Stitch's Great Escape) because guests under seven must be accompanied by an adult.
Fantasyland is where you should go first. You should remember to use your FASTPASS (FP) so you reduce wait times. Then head to Tomorrowland for Buzz Lightyear (FP) and Monsters, Inc. Comedy Club. Next stop Adventureland and Jungle Cruise (FP), Pirates of the Caribbean and Magic Carpets of Aladdin. Remember when you are waiting for your FP time to take advantage of the other attractions in the area.
If you have time and are not exhausted, head back to a favorite or schedule in the parade. Or if you want characters, go to Mickey's Toontown Fair. Or plan Advance Dining Reservations for characters at Crystal Palace with Winnie the Pooh and Friends.
